Maria de' Medici was the eldest legitimate daughter of Cosimo I de' Medici, Grand Duke of Tuscany and Eleonora di Toledo. She was a member of the famous Medici family.

Ferdinando II de' Medici was grand duke of Tuscany from 1621 to 1670. He was the eldest son of Cosimo II de' Medici and Maria Maddalena of Austria. He was remembered by his contemporaries as a man of culture and science, actively participating in the Accademia del Cimento, the first scientific society in Italy, formed by his younger brother, Leopoldo de' Medici. His 49-year rule was punctuated by the beginning of Tuscany's long economic decline, which was further exacerbated by his successor, Cosimo III de' Medici. He married Vittoria della Rovere, a first cousin, with whom he had two children who reached adulthood: the aforementioned Cosimo III, and Francesco Maria de' Medici, Duke of Rovere and Montefeltro, a cardinal.

Margherita de' Medici was Duchess of Parma and Piacenza by her marriage to Odoardo Farnese, Duke of Parma. Margherita was regent of Piacenza in 1635, and regent of the entire duchy from 1646 until 1648 during the minority of her son.

Garzia de' Medici was the son of Cosimo I de' Medici, Grand Duke of Tuscany, and Eleanor of Toledo. He was the subject of a famous painting by Bronzino when he was an infant. He was born in Florence and died of malaria along with his mother while traveling to Pisa, a few days after his brother, Cardinal Giovanni, also died of the disease.

Eleonor de' Medici was a Duchess of Mantua by marriage to Vincenzo I Gonzaga. She was a daughter of Francesco I de' Medici and Joanna of Austria. She was a family member of the famous House of Medici and the sister of Marie de' Medici the Queen of France.

The Portrait of Bia de' Medici is an oil-tempera on wood painting by Agnolo Bronzino, dating to around 1542 and now in the Uffizi in Florence. For a long time it was displayed in the Tribuna at the heart of the museum, but since 2012 it has been moved to the "sale rosse" of the Nuovi Uffizi. A second portrait, by Pontormo, has also been argued to show Bia de' Medici, but this identification is disputed.

Maddalena de' Medici (1473–1528) daughter to Lorenzo Medici Born in Florence, she was educated with her siblings to the humanistic cultures by figures such as Angelo Poliziano. In February 1487 she was engaged to be married to Franceschetto Cybo, son of Pope Innocent VIII. They were married in January 1488, and she brought a dowry of 4000 ducats. This marriage brought closer connections for her family and the Vatican, helping her brother Giovanni get appointed as a Cardinal. She used her influence with her father, her brother Piero, and the Pope to help friends and poorer people get aid and positions within the church and governments.

Caterina de' Medici was Duchess of Mantua and Montferrat as the second wife of Duke Ferdinando and Governor of Siena from 1627. She was the second daughter of Grand Duke Ferdinando I of Tuscany and his wife Christina of Lorraine.
